1. Silk Fibroprotein Reverses Fibroblast Senescence

In a UVB-induced senescent cell model, silk fibroin treatment reduced the number of cells positive for senescence markers (β-galactosidase), decreased SASP factor (IL-6, p16, p21) expression, accelerated cell proliferation, and restored the cell morphology from a "round and plump" shape to a "spindle-shaped" shape. After blocking the inflammatory pathway with the NF-κB inhibitor (BAY 11-7082), PLLA cells essentially lost their collagen-promoting ability (inflammation-dependent), while silk fibroin still stably promoted type I/III collagen secretion without increasing MMP-1/3 or showing p-p65 nuclear translocation. Histological sections showed fibrosis (high α-SMA expression) in the PLLA group, while the silk fibroin group showed almost no fibrosis.

Conclusion: Silk fibroin can directly awaken senescent fibroblasts, its action is independent of inflammation, and there is no risk of fibrosis.

2. Silk fibroin reduces collagen degradation

Transcriptomic/proteomic analysis identified the core pathway: ROS-MAPK (JNK)-AP-1 (c-Jun)-MMP (1/3). UVB leads to increased ROS → activation of JNK → upregulation of AP-1 → increased MMP → collagen degradation.

Photoaged nude mice: Silk fibroin inhibited MMP-1 and AP-1 overexpression and reduced collagenase activity.

Bama miniature pigs (structure most similar to humans): After 30 days of UVB irradiation, skin became rough, thickened, and collagen decreased. Silk fibroin increases type I/III collagen and elastin fibers, inhibits the MAPK/AP-1 pathway, and reduces MMP-1/AP-1. While PLLA increases collagen, it does not regulate the MAPK pathway.
