Reishi Extract : Hepatoprotective Activity and Mechanism of Action
A study published in *Scientific Reports*, a journal under Nature, investigated the alleviating effects of Ganoderma lucidum extract on oxidative stress and hepatotoxicity induced by carbon tetrachloride (CCL4) in Long Evans rats. The study indicated that Ganoderma lucidum extract possesses significant hepatoprotective function, with its mechanism of action primarily manifested in reducing oxidative stress and inflammatory responses by inhibiting the expression of pro-inflammatory cytokines and promoting the expression of the anti-inflammatory factor IL-10.
To verify the protective effect of Ganoderma lucidum against CCL4-induced hepatotoxicity in rats, researchers divided the rats into five groups. Four groups were exposed to CCL4. One group received additional silymarin (a known hepatoprotective component), two groups were treated with Ganoderma lucidum extract at doses of 100 mg/kg and 200 mg/kg, respectively, and the remaining group received no intervention as a control.
Experimental Results
Result 1: Enhanced In vivo antioxidant enzyme levels
Compared to rats treated with CCL4 alone, rats treated with both CCL4 and Ganoderma lucidum extract showed increased plasma levels of glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT), indicating that Ganoderma lucidum extract can enhance the activity of antioxidant enzymes in vivo.
Result 2: Inhibition of inflammatory response
The expression of the pro-inflammatory factor IL-1β was detected in rats treated with CCL4 alone, but this expression was inhibited after combined treatment with Ganoderma lucidum extract. Simultaneously, the expression of the anti-inflammatory factor IL-10 was suppressed in the CCL4-treated group, but the expression level of IL-10 was significantly increased in the Ganoderma lucidum extract-treated group. IL-10 can inhibit specific immune functions of macrophages and reduce the release of inflammatory factors, thereby exerting an anti-inflammatory effect.
Result 3: Alleviation of Hepatotoxicity
Compared to the untreated group and the group treated with Ganoderma lucidum extract, rats treated with CCL4 alone showed higher levels of TNF-α expression. Furthermore, CCL4-induced hepatotoxicity also led to increased expression of transforming growth factor-β (TGF-β), a factor associated with liver failure. However, in the group treated with Ganoderma lucidum extract, TGF-β expression decreased. These results indicate that Ganoderma lucidum extract helps alleviate hepatotoxicity.
Conclusion
This study demonstrates that Ganoderma lucidum extract has significant hepatoprotective activity. The mechanism involves inhibiting the expression of pro-inflammatory cytokines and promoting the expression of anti-inflammatory factors such as IL-10, thereby reducing oxidative stress and inflammatory damage.Ganoderma lucidum has the potential to be developed into a hepatoprotective agent and is a promising ingredient.
