Introduction

It is a proteolytic enzyme extracted from bovine or porcine pancreas. It is white or off-white crystalline or amorphous powder, soluble in water and insoluble in organic solvents. It is mainly secreted by the pancreas and activated by trypsin. As a digestive enzyme that can decompose proteins, the active group is serine, so it belongs to serine protease, which cuts peptide bonds at the carboxyl group of tyrosine, tryptophan and phenylalanine, thereby cleaving proteins. Purification Process

(1) Take the bovine pancreas, remove the fat and connective tissue, and mince it into pancreatic pulp;

(2) Add sulfuric acid solution to the pancreatic pulp for extraction and filter;

(3) Add ammonium sulfate to the filtrate to 0.4 saturation and place it in a cold room overnight;

(4) Filter, and ultrafilter the filtrate with an ultrafiltration membrane with a molecular weight cutoff of 50kDa;

(5) Ultrafilter the filtrate with an ultrafiltration membrane with a molecular weight cutoff of 5-10kDa;

(6) Add ammonium sulfate to the retentate to 0.7 saturation and place it at low temperature. Leave overnight;

(7) Filter, dissolve the filter cake in cold purified water, the volume of cold purified water is 3-5 times the mass of the filter cake, add sulfuric acid to adjust the pH to 2.0, stir to dissolve, add saturated ammonium sulfate solution, the volume of saturated ammonium sulfate solution is 2 times the mass of the filter cake, adjust the pH to 5.0, 25℃, incubate for 2-5h, crystals precipitate, filter and collect the crystals;

(8) Dissolve the crystals with cold purified water and sulfuric acid, the volume of cold purified water is 3-5 times the mass of the crystals, adjust the pH 7.6, add trypsin, activate at 20℃ for 2-4h;

(9) Adjust pH to 4.0, add solid ammonium sulfate to reach 0.7 saturation, filter and collect precipitate;

(10) Dissolve the precipitate with cold purified water and sulfuric acid, and ultrafilter with an ultrafiltration membrane with a molecular weight cutoff of 10kDa;

(11) Add the above ultrafiltration desalted liquid to the SP ion exchange column that has been equilibrated with 0.05mol/L NaAC buffer, wash the column with 3 column volumes of buffer containing 0.2 mol/L NaCl, and elute with 3 column volumes of buffer containing 0.4 mol/L NaCl to obtain eluate;

(12) Ultrafilter the above eluate with an ultrafiltration membrane with a molecular weight cutoff of 10-30kDa, sterilize, and obtain.
