What is Glucose Oxidase?
Glucose Oxidase Powder, referred to as GOD, is a protein with a molecular weight of about 160,000 Daltons, composed of two (or four) polypeptide chains. The optimum pH of the enzyme reaction is 5-7, and the optimum temperature is 30-40°C. Flavin adenine dinucleotide (FAD) is required as a coenzyme to participate in the catalysis.
GOD is an aerobic dehydrogenase, belonging to the oxidoreductase class. Under aerobic conditions, β-D-glucose can be specifically oxidized by glucose oxidase into D-gluconic acid-δ-lactone and hydrogen peroxide, and D-gluconic acid-δ-lactone is spontaneously hydrolyzed into gluconic acid.
In the body, it plays an important physiological function, such as regulating blood sugar levels and energy metabolism. In addition, it also has a wide range of application value, such as playing an important role in food, medicine and other fields.
Characteristics of GOD
High-purity GOD is a light yellow powder, easily soluble in water, with a molecular weight of about 150KD and light absorption wavelengths of 377nm and 455nm. The action temperature of GOD is 30℃~60℃, and the solid enzyme preparation can be stable for at least 2 years at 0℃ and 8 years at -15℃. The stable pH range of GOD is 4.0~8.0, and it has good stability. Generally speaking, enzyme reactions are not greatly affected by substrate concentration. When the glucose concentration is 5%~20%, the reaction rate remains almost unchanged.
Physiological Function
Catalyze glucose oxidation: It can specifically catalyze the reaction of β-D-glucose with oxygen to produce gluconic acid and hydrogen peroxide. This reaction helps the metabolism and utilization of glucose in the body.
Antioxidant: By consuming oxygen and generating hydrogen peroxide, it plays an antioxidant role in food and feed, protects easily oxidized components, and prolongs their shelf life.
Improve intestinal environment: It can improve the intestinal environment of animals, promote the growth of beneficial bacteria, inhibit harmful bacteria, and thus improve the digestion and absorption capacity and immunity of animals.
Anti-mold: It can catalyze the oxidation of β-D-glucose into D-glucono-δ-lactone and hydrogen peroxide in an aerobic environment. This process consumes oxygen, thereby inhibiting the growth of aerobic bacteria, and reducing the growth of mold and the production of mycotoxins, extending the shelf life of food and feed.
Protect intestinal epithelial cells: By eliminating free radicals, it protects the integrity of intestinal epithelial cells and prevents the invasion of pathogens.
Promote digestion and absorption of nutrients: It can increase the acidity of the intestine, maintain the activity of proteases, promote the absorption and utilization of nutrients such as amino acids and fatty acids, and improve feed utilization.
Mechanism of Action
GOD can consume oxygen to catalyze glucose oxidation, producing hydrogen peroxide and gluconic acid.
1. The enzymatic reaction of GOD is not greatly affected by the substrate concentration. The reaction rate is almost unchanged when the glucose concentration is 5%~20%.
2. According to the optical rotation measurement, the initial product of the GOD reaction is δ-gluconolactone, which is spontaneously hydrolyzed to gluconic acid by non-enzymatic reaction.
3. Na+, Ca2+, Mg2+, Zn+ and Mn+ have different degrees of activation effects on the enzyme. GOD is only active on D-glucose and has no activity on L-glucose.
4. In the presence of catalase, catalase can decompose hydrogen peroxide to produce water and oxygen, and water combines with gluconolactone to produce gluconic acid.
