1. Chromatographic conditions

Preparative HPLC conditions: self-filled Microsorb C18 column (50 mm X200 mm, 5μm); mobile phase: methanol water (volume ratio of 60:40), flow rate 100mL/min; detection wavelength: 220 nm; injection volume: 1.5mL.

Analytical HPLC conditions: Microsorb C18 column (4.6mmX250mm, 5μm) filled by Dalian Jiangshen Company; mobile phase: methanol water (volume ratio of 60:40), flow rate 1mLmin; detection wavelength: 200 nm; injection volume: 10μL.

2. Experimental method

Weigh 6g of crude Centella asiatica glycoside sample and dissolve it in methanol to prepare a solution with a concentration of 40mg/mL, pass through a 0.45μm microporous filter membrane for sample analysis and preparation. Take an appropriate amount of the treated sample solution, dilute it 10 times with methanol and analyze it. Take asiaticoside and madecassoside as reference substances and quantify it by external standard method. The crude asiaticoside sample contains 20% asiaticoside and 45% madecassoside. Prepare and purify it by preparative HPLC system, inject it multiple times, collect the eluent containing asiaticoside and madecassoside, concentrate it under reduced pressure, and dry it in an oven to obtain 0.8g asiaticoside and 2g madecassoside.
