Preparation Method Of S-Acetyl-L-Glutathione

Acetyl chloride (1.4 kg, 17.83 mol) was slowly added to a solution of L-glutathione (5.0 kg, 16.27 mol) in the presence of trifluoroacetic acid (15 L), DMF (15 L) and CoCl2 (10.56 g, 0.081 mol) at 0-5°C in a dry reaction flask for 5 minutes. After the addition was complete, the reaction temperature was raised to 20-30°C and the resulting reaction mixture was stirred for 10 minutes. The progress of the reaction was monitored by HPLC. After the reaction was completed, 0.21 L of methanol was slowly added to the above reaction mixture to quench the reaction and the reaction mixture was stirred for another 20 minutes at room temperature. TFA and DMF were removed in vacuo and ethyl acetate (50 L) was added to the resulting oil. The resulting reaction mixture was cooled to 0°C, and the crude precipitate was dissolved in warm water (40°C) and the resulting solution was diluted with 60 L of acetone. The mixture was cooled in an ice-water bath for about 2 hours, and the formed white crystalline substance was collected to obtain the target product molecule S-acetyl-L-glutathione.
