What Is Tetrahydropalmatine?
Tetrahydropalmatine (THP) is an alkaloid extracted from the dried tuber of Corydalis yanhusuo, a plant of the genus Corydalis of the family Papaveraceae. However, it has poor water solubility, low solubility, is easily metabolized in the body, and its oral absorption bioavailability is only 6.59%. It is highly soluble in ether, chloroform, soluble in ethanol, and almost insoluble in water or alkaline solutions. Its hydrochloride and sulfate are both soluble in water and are racemic. The effective one is the levorotatory isoform, named cranial pain dine. These physical and chemical properties limit the wide application of tetrahydropalmatine. It has multiple pharmacological activities such as sedation, analgesia, anti-myocardial ischemia, anti-thrombosis, inhibition of gastric acid secretion, anti-arrhythmia, and myocardial ischemia/necrosis protection.

A New Mechanism of Tetrahydropalmatine(THP)
Angiogenesis is the process of endothelial cells forming new blood vessels. It is an adaptive response of vascular endothelial growth factor (VEGF) to nutrients or hypoxia during exercise or ischemia. THP is the index component of THP and has significant sedative, hypnotic and analgesic effects. It can block calcium channels, inhibit pathogens and conditioned reflexes, and protect brain tissue and cardiovascular system.
Through pharmacological experiments and DESI-MSI spatial metabolomics and GC-MS non-targeted metabolomics, it was identified as a trigger factor for promoting angiogenesis in chicken embryo chorioallantoic membrane in vivo and multiple endothelial cell models in vitro.

At the same time, THP significantly reduced t-BHP-induced oxidative stress.
THP is a pro-angiogenic trigger by regulating arginine biosynthesis. It can sequentially affect the flow of citrulline to arginine, arginine biosynthesis and endothelial VEGFR2 expression, thereby promoting angiogenesis. These results indicate that THP is a potential pro-angiogenic lead compound.
