Aging is a key risk factor for numerous age-related diseases, such as type 2 diabetes, osteoporosis, Alzheimer's disease, and cancer, placing a significant burden on society. Therefore, exploring methods to reduce the incidence of these diseases and improve overall health is of paramount importance.

Bile acids are naturally occurring metabolites and the main component of bile, primarily concentrated in the enterohepatic circulation system. They can be divided into primary and secondary types and play a central role in regulating lipid and glucose metabolism, as well as other metabolic processes.

Previous studies have shown that a secondary bile acid called lithocholic acid (LCA) can extend the lifespan of yeast. TUDCA (tauroursodeoxycholic acid) powder is also a naturally occurring secondary bile acid, synthesized in the liver by combining UDCA (ursodeoxycholic acid) with taurine. However, the specific effects of TUDCA on lifespan and healthspan have been unclear.

On September 24, 2024, a study from the Nanhu Laboratory of the National Center for Biomedical Analysis was published in the journal Science China Life Sciences. The paper, titled "Tauroursodeoxycholic acid targets HSP90 to promote protein homeostasis and extend healthy lifespan," demonstrated that supplementation with TUDCA significantly extended the lifespan of nematodes and improved their health markers.

To investigate whether bile acids have lifespan-extending effects, the researchers tested the effects of various bile acids on the lifespan of Caenorhabditis elegans. They found that only TUDCA significantly extended lifespan. TUDCA Powder was administered continuously from the L4 larval stage until death. Treatment with TUDCA at different concentrations (2.5, 10, and 50 mmol/L) extended the average lifespan of the nematodes by 8.70%, 23.72%, and 19.12%, respectively.

The research team further evaluated the effects of TUDCA on the health of the nematodes. Data showed that TUDCA treatment significantly improved nematode motility compared to the control group, as evidenced by increases in bending frequency and pharyngeal pumping rate. A key finding was that the lifespan-extending effect of TUDCA did not come at the expense of reproductive capacity. Furthermore, the study observed that TUDCA ameliorates aging-related phenotypes in mice.

Mechanistically, the researchers discovered that TUDCA supplementation triggered widespread transcriptional changes in genes associated with anti-aging traits. The molecular mechanism is that TUDCA targets the molecular chaperone protein HSP90, enhancing its protein refolding function. This action, in turn, alleviates aging-induced endoplasmic reticulum (ER) stress, promotes the maintenance of protein homeostasis, and thus enhances resistance to aging.

In summary, this study elucidates the molecular link between endogenous metabolites and protein homeostasis, proposes a novel anti-aging strategy, and opens new avenues for extending lifespan and healthspan.

How Should TUDCA Powder Be Taken?

A variety of dosages have been studied for the benefits of TUDCA. Starting with a TUDCA supplement of 10-13 mg per day, patients with chronic liver disease experienced significant reductions in liver enzymes for three months. Doses of up to 1,750 mg per day have been shown to benefit fatty liver disease and improve muscle and liver insulin sensitivity. Animal studies have shown that doses up to 4,000 mg (the human equivalent) have a positive impact on neuroprotection against age-related memory loss.

Despite these extreme dosages, between 500 mg and 1,500 mg per day appears to be an ideal dose to produce the effects of TUDCA. Most supplements appear to be formulated to contain 100–250 mg of TUDCA per serving, to be taken multiple times daily. As with many of these ingredients, more research is needed to obtain some specific figures.

When should it be taken?

TUDCA can be taken at any time of day, preferably with food to aid absorption. As mentioned above, most supplements have a dosage of 100–250 mg per serving. It's recommended to spread TUDCA dosages throughout the day, taking them 2, 3, 4, or even 5 times daily.

How long does it take for the effects to be felt?

TUDCA powder doesn't work overnight. Studies have shown varying effects after 1, 2, 3, or even 6 months of supplementation. Based on available research, it's safe to assume that at least 30 days (1 month) of supplementation are necessary to see improvements and benefits. However, consistent and long-term use will yield the greatest benefits from TUDCA supplementation.
