Effect on FLS cell viability
CCK-8 assay was used to detect its effect on FLS cell viability. FLS cells were treated with different concentrations for 24h. When the concentration was ≥80μmol/L, the viability of FLS cells was significantly reduced, and with the continuous increase in concentration, the cell survival rate decreased in a dose-dependent manner, indicating that this concentration had a significant inhibitory effect on the survival rate of FLS cells.
Effect on the proliferation of activated FLS cells
Under the pathological state of RA, FLS is abnormally activated and proliferates abnormally. TNF-α (10ng/mL) was used to activate normal FLS to turn it into RA-FLS. Compared with the blank group, the FLS in the model group was activated, showing significant proliferation of RA-FLS. It shows that tripterygium wilfordii can significantly inhibit the proliferation of RA-FLS and inhibit the progression of RA at 10-40μmol/L.
Effects on inflammatory cytokines and matrix metalloproteinases in activated FLS
When its concentration was 40μmol/L, it could significantly inhibit the release of inflammatory cytokines (P<0.000 1, P<0.001). In the range of 10~40μmol/L, it could significantly inhibit the production of related matrix metalloproteinases (MMP-1 and MMP-3).
Effects on the expression of apoptotic proteins in activated FLS
After 2h of treatment, the protein expression level of the high-dose group (40μmol/L) was significantly increased.
Effects on ERK1/2 and phosphorylated ERK1/2 in activated FLS
Compared with the TNF-α model group, both showed activation of phosphorylated ERK1/2 protein.
Conclusion, the mechanism of action of inhibiting TNF-α-induced rheumatoid arthritis may be related to its inhibition of the activation of phosphorylated ERK-related pathway proteins and the promotion of apoptosis of RA-FLS.
