An experimental study investigated the effects and mechanisms of rosemary essential oil on physical fatigue (caused by excessive exercise) and central nervous system fatigue (caused by sleep deprivation), providing a scientific basis for the development of anti-fatigue health products.
Experimental Methods
Mental alertness and anti-hypoxia: Mice were divided into a control group, a short-acting inhalation group (2 hours daily), and a long-acting inhalation group (12 hours daily), and inhaled 0.5% rosemary essential oil for 7 days via aromatherapy. Subsequently, spontaneous activity, sodium pentobarbital sedation-hypnosis, hypoxia tolerance, rotarod tests, and weighted swimming tests were conducted.
Mechanism of resistance to physical fatigue: Serum urea nitrogen, lactate, lactate dehydrogenase, muscle/liver glycogen, oxidative stress markers (SOD, MDA), and key proteins of the Nrf2/ARE pathway in skeletal muscle (Keap1, Nrf2, NQO1, HO-1) were measured.
Mechanism of resistance to central nervous system fatigue: A central nervous system fatigue model was established through 14 hours of mechanical sleep deprivation daily for 3 weeks, while simultaneously inhaling essential oil for 3 hours daily. Y-maze and open field experiments were performed to detect hypothalamic neurotransmitters (5-HT, DA), hippocampal neuronal morphology was observed, and hippocampal Sirt1/PGC-1α/Nrf1 pathway proteins were detected.
Component analysis: GC-MS was used to identify the essential oil components, and the main components were molecularly docked with the target sites Nrf2 and Sirt1.

Results
Mental alertness and anti-hypoxia effects: The essential oil group showed significantly increased spontaneous activity, standing frequency, hypoxia survival time, rotarod time, and swimming time; sodium pentobarbital-induced sleep duration was significantly shortened.
Anti-physical fatigue mechanism: Serum urea nitrogen, lactate, and lactate dehydrogenase activity were significantly decreased in the essential oil group; muscle/liver glycogen content and skeletal muscle SOD activity were significantly increased; MDA content was significantly decreased. Simultaneously, Keap1 protein expression was decreased in skeletal muscle, while Nrf2, HO-1, and NQO1 protein expression were increased.
Anti-central fatigue mechanism: Mice in the essential oil group showed a significantly increased spontaneous alternation rate in the Y-maze (improved cognition); significantly reduced time spent in the central region of the open field (relieved anxiety); and increased total distance traveled. Hypothalamic 5-HT levels were significantly decreased, while DA levels were significantly increased. Expression of Sirt1, PGC-1α, Nrf1, and TFAM proteins in the hippocampus was significantly upregulated.
Active components: The main components of the essential oil include isoborneol acetate, 2-carene, camphor, 1,8-cineole, and dextrorotatory borneol. Molecular docking showed that camphor, isoborneol acetate, and dextrorotatory borneol had strong binding affinity to Nrf2 and Sirt1.
Conclusion
According to the study, rosemary essential oil has the effects of refreshing the mind, resisting hypoxia, and combating fatigue. Its anti-physical fatigue mechanism is through activating the Nrf2/ARE signaling pathway in skeletal muscle, while its anti-central fatigue mechanism is through activating the Sirt1/PGC-1α/Nrf1 signaling pathway in the hippocampus to promote mitochondrial biosynthesis, thereby improving cognitive impairment and anxiety. Camphor, isoborneol acetate, and dextrorotatory borneol are its main active ingredients.
