N-acetyl-L-cysteine ​​(NAC) is the acetylated form of L-cysteine, a sulfur-containing amino acid that the human body can synthesize, but under certain conditions, its synthesis is insufficient to meet requirements. Therefore, NAC is considered a semi-essential amino acid; NAC supplementation is more easily tolerated by the body, has higher bioavailability, and lower susceptibility to oxidation.

NAC: Can It Be Used In Feed?

N-acetyl-L-cysteine ​​(NAC), as a natural ROS scavenger, is increasingly being cited as a feed additive to enhance the antioxidant activity of aquaculture species.

Studies have shown that N-acetyl-L-cysteine ​​(NAC) and lauric acid (LA) provide effective antioxidant and antibacterial feed additives for high-density cultured Litopenaeus vannamei. This study evaluated whether LA and NAC can be used as feed additives to improve the immune and antioxidant responses of Litopenaeus vannamei cultured at high densities.

The study consisted of five groups:

1: Low-density culture + basal feed (negative control)

2: High-density culture + basal feed (positive control)

3: High-density culture + supplemented with 0.2% lauric acid (LA)

4: High-density culture + supplemented with 0.2% NAC

5: High-density culture + supplemented with 0.2% LA + 0.2% NAC

The experiment lasted for eight weeks, primarily examining the growth performance (body weight, feed conversion ratio), blood immune indicators (total blood cell count, phagocytic activity, total protein, etc.), antioxidant indicators (MDA, SOD, GPX), and the expression of immune-related genes (LYS, PPOD) in shrimp.

Conclusion

The results showed that under high-density culture conditions, the addition of NAC, LA, or a combination of both significantly improved the health status of Litopenaeus vannamei.

The NAC group (4) showed the best overall performance, with the highest body weight gain and the most significant antioxidant effect, effectively reducing the level of the oxidative stress marker MDA and improving antioxidant capacity. LA significantly enhances antibacterial and immune functions, increasing lysozyme (LYS), prophenol oxidase (PPOD), hemocyte count, and phagocytic activity, thus contributing to improved innate immunity against bacteria and viruses.

The combined use of LA and NAC (5) provides both antioxidant and immune-enhancing effects, showing superior performance in some immune indicators compared to individual additions.

Adding LA and NAC can also improve nutrient utilization and gut health, increasing total protein and glucose levels, thereby promoting growth.

Overall, 0.2% NAC and 0.2% lauric acid, as feed additives, can effectively improve the growth performance, antioxidant capacity, and immunity of high-density cultured Litopenaeus vannamei, demonstrating good potential for aquaculture applications. However, researchers believe further studies on pathogen challenge experiments and different dosages are needed to determine the optimal application method in commercial aquaculture.
